摘要
目的:探讨肢体缺血再灌注(limbs ischemia-reperfusion,LIR)对脑组织的影响。方法:复制大鼠LIR模型,采用尼氏染色观察脑组织病理学改变,TUNEL法检测细胞凋亡情况,免疫组化和Western印迹法检测凋亡相关因子Bcl-2、细胞色素C(cyto C)、Caspase-3的表达变化。结果:大鼠LIR后,脑组织海马区、中脑红核区神经元细胞受损,脑组织出现水肿裂隙,cyto C、 Caspase-3凋亡相关蛋白表达明显上调,24 h达高峰.与细胞凋亡数量的增加相一致,Bcl-2表达与再灌时程呈负相关。结论:LIR 后可导致脑组织损伤,并出现细胞凋亡,其发生机制与Caspase-3、cyto C、Bcl-2凋亡相关蛋白的表达变化有关。
Objective: To evaluate the effects of hind limbs ischemia-reperfusion (LIR) on brain injury, Methods: Limbs ischemia- reperfusion model was established in rats and the histo-pathological changes were observed by Nissl staining. Apoptotic cells were detected with TUNEL and the expressions of the associated proteins, such as Bcl-2, cyto C and caspase-3 were determined with immunohistochemistry and Western blot. Results: The expressions of Caspase-3 and cyto C following LIR were significantly increased compared with those in the control group (P〈 0. 05) and peaked 24 h later. The expression of Bcl-2 was negatively associated with reperfusion time, The expressions of Caspase-3 and Bcl-2 gene were obvious in the hippocampus and the midbrain red nucleus region, which was the sensitive area to ischemic injury. Conclusion: Our results indicate that LIR can lead to apoptotic brain injury. Bcl-2, cyto C and Caspase-3 may play a vital role in regulating the neural apoptosis following LIR in rats.
