摘要
基于脱氧核糖核酸(DNA)对有机染料碱性品红的共振光散射增强效应,拟订了一种测定DNA的共振光散射法.在pH=6.75~7.25的范围内,碱性品红在594nm处的共振光散射增强与yDNA和ctDNA的浓度呈线性关系,线性范围分别为0.20~1.60 μg/mL和0~1.50μg/mL,相关系数分别为0.999 7和0.999 4,检出限可达26.5 μg/L.该方法简便、快速,用于合成样品中DNA的测定,结果满意.
The resonance light-scattering(RLS) of fuchsin basic is greatly enhanced by deoxyribonucleic acid(DNA) in the pH range of 6.75~7.25. The enhanced intensity of RLS at 594 nm is proportional to the concentration of DNA in the range of 0.20~1.60μg/mL with a correlation coefficient of 0. 9997. The detection limit is 26. 5μg/L. This method is simple and rapid. It has been applied to the determination of DNA in synthetic samples with satisfactory results.
