摘要
介绍在阳离子表面活性剂十六烷基三甲基溴化铵 (CTMAB)存在下阴离子染料铬天青S(CAS)共振光散射(RLS)法测定脱氧核糖核酸 (DNA)的方法。在pH =5 .2 7的六次甲基四铵 -盐酸缓冲溶液中 ,研究了CAS -CTMAB -yDNA体系的RLS光谱特征、影响因素和最佳反应条件。在最佳条件下 ,体系的RLS强度增加值ΔI与yDNA的浓度在 5 0~ 80 0 μg/L和 10 0 0~ 2 0 0 0 μg/L呈线性关系 ,其线性回归方程分别为ΔI=0 .4 8c+2 .5 6和ΔI=0 .14c +17.86 ,相关系数分别为 0 .9997和 0 .9991,检出限为 4 .3ng/mL。该方法简便、快速 ,应用于合成样品中yDNA的测定 ,测定结果的相对标准偏差为 2 .93%~ 4 .71% ,回收率为 97.8%~ 10 5 .2 %。
The determination of deoxyribonucleic acid with chrome azurol S by resonance light scattering method in the presence of cation surfactant cetyltrimethylammonium bromide (CTMAB)was reported. In hexamethylene tetramine-HCl buffer solution(pH=5.27), the spectral characteristics of RLS, the affecting factors and the optimum conditions of the reaction were investigated. Under the optimum conditions, the enhanced RLS intensity is proportional to the concentration of yDNA in the range of 50~800 μg/L and 1000~ 2000 μg/L. The linear regression equation are Δ I=0.48c+2.56 and Δ I=0.14c+17.86 respectively, the correlation coefficient are 0.9997 and 0.9991 respectively and the detection limit is 4.3 ng/mL .This method is simple,rapid and was applied to the determination of yDNA in synthetic samples with the recoveries of 97.8 ~105.2% and the relative standard deviations of 2.93%~ 4.71% .
