摘要
麦角菌[Clavicepspurpurea(Fr.)Tul.ATCC20019]发酵生产麦角隐亭的最佳碳源是蔗糖或工业葡萄糖,蔗糖的最适浓度为30%,麦角隐亭产量为97.8—109.3μg/mL;发酵前期在10%蔗糖培养基中再补加20%蔗糖,可使产碱量比直接用30%蔗糖的产碱量增加到111.9μg/mL.最佳氮源是酵母粉、蛋白胨、蚕蛹粉和黄豆饼粉.正交L16(42×29)证明,硫酸镁是10种发酵条件中对该菌株产麦角隐亭的量影响最大的一种.
Ergocryptine is produced by Claviceps purpurea (Fr.) Tul. ATCC 20019 in submerged liquid culture under aerobic condition. Sucrose and industrial glucose are the suitable carbon sources when citric acid is present. The suitable sucrose concentration is 30% and the amounts of alkaloid are 97.8-109.3μg/mL. Adding 20% sucrose again to a liquid culture containing 10% sucrose at the earlier fermentation period can enhance alkaloid production. The most favourable nitrogen sources selected are yeast extract, peptone, silkworm chrysalis and soybean cake powder. It is demonstrated that magnesium sulphate is the most important effective factor for alkaloid production by latin square L16(4 2×2 9).
