摘要
目的单细胞凝胶电泳技术是检测细胞DNA损伤与修复的一种快速、简便、灵敏方法,但影响因素很多,为此对该技术进行了改进,并对操作中易出现的问题进行了总结。方法在singh等人的方法基础上,根据本实验室条件进行改进,以获得更好、更稳定的结果。结果改进后技术检测H2O2对K562细胞损伤具有良好的剂量效应关系。结论改进方法不影响结果的正确性,更易于掌握和操作,减少了影响因素,试验更具实用性,可以广泛应用于离子射线损伤、自由基损伤、癌症放化疗、生物监测等领域。
Objective: To improve the technology and summary the problems of single-cell gel electrophoresis in detecting the damage and repair of cell DNA. Methods: On basis of the method of the Singh, an improvement on this technology was made according to the conditions of our experiment in order to achieve better and more stable results. Results: The improved method of detecting H2O2 vs K562 damage has a good dosage-domino effect. Conclusion: The improved method does not affect the validity of the result, and is easier to be mastered and processed. It also reduces affecting factors and makes the experiments more practicable.
