摘要 目的探讨甲氧滴滴涕(Methoxychlor,MXC)对雄性大鼠生精细胞的细胞周期影响。方法用流式细胞仪检测染毒雄性大鼠生精细胞周期变化。结果随着MXC染毒时间延长,实验组G0/G1期细胞数(5、10和15 d组分别为58.4%、45.8%和43.9%)与S期细胞数(5、10和15 d组分别为21.9%、20.8%和12.7%)减少,G2/M期细胞数(5、10和15 d组分别为19.7%3、3.4%和43.4%)增多(P<0.05);实验组单倍体细胞减少,二倍体与四倍体细胞增多(P<0.05);除5 d实验组的二倍体与四倍体细胞平均荧光强度增强外,其余实验组单倍体、二倍体和四倍体细胞平均荧光强度减弱(P<0.05),溶剂组变化差异无显著性(P>0.05)。结论MXC可以引起大鼠睾丸生精细胞S时相抑制,并出现G2期细胞阻滞。 Objective To investigate the effect of methoxychlor(MXC) on cell cycle of germ cells in male rat. Methods The cycle change of germ cells was examined with Flow Cytometry in rat exposed to MXC. Results The cell number of G0/G1 (58.4%, 45.8% and 43.9% on 5,10 and 15 d separately) and S phase (21.9% ,20.8% and 12.7% on 5,10 and 15 d separately) decreased, but that of G2/M phase (19.7% ,33.4% and 43.4% on 5,10 and 15 d separately) increased in experimental group with extension of MXC exposure( P 〈 0.05). The number of hapoloid cell decreased, number of diploid and tetraploid cell increased in experimental group (P 〈 0.05). The mean fluorescence intensity of diploid and tetraploid cell elevated in 5 days experimental group, but that decreased in other experimental groups ( P 〈 0.05). The change was no difference in solvent group ( P 〉 0.05 ). Conclusion MXC can inhibit S phase of germ cell and block cell in G2 phase in rat spermary.
