摘要
根据已报道的猪抗菌肽PR-39基因和看家基因β-actin的序列,设计了针对这两种基因的引物,构建了以MgC l2浓度为1.5 mmol/L,循环次数为29的优化半定量RT-PCR法。以β-actin为内标,研究了不同生长阶段猪抗菌肽PR-39基因的表达差异。结果表明,从1日龄到7周龄,PR-39基因表达呈上升的趋势;在7周龄到14周龄,PR-39基因表达略有下降;14周龄到21周龄,PR-39基因表达又有上升的趋势;21周龄到28周龄基因表达再次下降。
The primer for PR-39 and β-actin was designed respectively according to the reported porcine gene sequence. The results indicated that, to ensure the feasibility and reliability of semi-quantitative RT-PCR method, the suitable MgCl2 concentation and cycles in PCR should be 1.5 mmol/L and 29 respectively. An optimized semi-quantitative RT-PCR was constructed to detect the PR-39 gene expression difference at different growth stages of swine by using β-actin as inner control. The results showed that PR-39 gene expression increased from one day to 7 weeks, decreased from 7 to 14 weeks, increased from 14 to 21 weeks and reduced again at the stage of 21 to 28 weeks.
