摘要
目的:研究哮喘豚鼠气道内神经激肽1受体(NK-1R)mRNA的分布及相对含量。方法:用卵蛋白超声雾化法制作哮喘豚鼠模型;原位杂交技术检测NK-1R mRNA,光镜观察其在气道内表达的部位。图像分析测定NK-1R mRNA的相对含量。结果:哮喘组NK-1R mRNA表达程度明显高于正常组;NK-1RmRNA分布于气管上皮细胞上层细胞表面、黏膜下层、血管周围上皮细胞、炎性细胞的表面和平滑肌细胞、腺体细胞表面。结论:NK-1R参与了卵蛋白诱导的哮喘豚鼠发病机制。
Objective: To study the expression and content of neurokinin-1 receptor(NK-1R) mRNA in the airway of asthmatic guinea pigs. Methods :The model of asthmatic guinea pigs was established by aerosolized ovalbumin. The expression of NK-1R mRNA was detected by in situ hybridization, and the expression site was observed under the light microscope. The content of NK-1R mRNA was determined by image analysis. Results:The expression of NK-1 R mRNA in asthma group was higher than that in normal control group. NK-1R mRNA was expressed on the cell surface of tracheal epithelia, submucosa, blood vessels epithelia, inflammatory cells, smooth muscle ceils, and gland ceils. Conclusion:NK-1R may be involved in the pathogenesis of ovalbumin-induced asthma in guinea pigs.
