高血压局灶性脑缺血大鼠脑细胞膜磷脂水平变化

摘要 目的探讨高血压局灶性脑缺血对脑细胞膜磷脂代谢的影响。方法采用 SD 大鼠慢性肾血管性高血压、大脑中动脉阻断脑缺血模型,高效液相色谱定量法测定高血压组(n=7)、高血压脑缺血组、正常血压脑缺血组缺血1,5,15,60,360 min脑细胞膜磷脂组分变化。结果高血压脑缺血组磷脂酰肌醇在缺血早期显著低于高血压组(n=7)(P<O.05~0.01),磷脂酰乙醇胺、磷脂酰胆碱分别在缺血15 min、60 min 始显著低于高血压组(P<0.01~0.05)。高血压脑缺血组与正常血压脑缺血组比较显示,前者磷脂酰肌醇(缺血1~5 min)、磷脂酰丝氨酸(缺血5 min)、磷脂酰乙醇胺(缺血15,360 min)、磷脂酰胆碱(缺血60~360 min)水平显著低于后者(P<0.05).结论高血压局灶性缺血早期首先出现脑细胞膜功能磷脂降解,膜结构磷脂则在缺血后期出现降解;高血压、脑缺血因素共同作用下将加重脑细胞膜的损害程度。 Aim To Determine the changes of neuronal membrane phospholipid content in focal cerebral ischemic in rats.Methods The content of phospholipids(PLs)of the cerebral neuronal mem- brane were determined in hypertensive focal cerebral ischemia model of Sprague-Dawley rats(n=7),after different time of ischemia(1,5,15,60,360min)us- ing high-performance liquid chromatography tech- nique.Results In the hypertensive cerebral is- chemic group phosphatidylinositol(PI)in cerebral neronal membrane was remarkable reduced from ear- lier stage of ischemia in comparison with hypertensive group(P<0.01~0.05),but the most prominent de- creases in phosphatidylethanolamine(PE),phos- phatidylcholine(PC)were found in latter stage of is- chemia(15~360min)(P<0.01~0.05).The levels of PI(1,5min),phosphatidylserine(PS)(5min),PE (15,360min),PC(60~360min)in hypertensive cerebral ischemic group were significantly lower than that of normotensive cerebral ischemic group(P< 0.05).Conclusion The PLs of and the membrane structural were decomposed during the early period of ischemia.The functional phospholipid in neuronal membrane was decomposed at the earlier stage of cerebral ischemia,while the structure phospholipid in neuronal membrance was decomposed at the late stage.Hypertension associated cerebral ischemia ag- gravated the damage of cerebral cell membrane.
