实验性脑出血大鼠急性期不同脑区及心肌内皮素mRNA的动态表达 被引量:1

Dynamic expression of ET mRNA on different cerebral region and myocardium at acute stage of intracerebral hemorrhage in experimental rats

摘要 目的探讨内皮素(ET)在脑心综合征发病机制中的作用。方法利用组织原位杂交技术检测实验性脑出血大鼠的中枢心血管特定调节区域及心肌ETmRNA表达的动态变化,并对阳性反应物进行图像定量分析。结果实验性脑出血可迅速诱导ET基因在脑出血周围区、下丘脑、脑干、海马、心肌的异常表达。脑出血6h即可见ETmRNA表达上调,到24h达高峰,72h时段虽略有减少,但仍高于正常水平。同时心肌的ET表达增多。结论脑出血周围区及中枢心血管调节区域的ET过多释放,这可能是脑心综合征的病理基础之一。 Objective To explore the effect of endothelin (ET) in pathogenic mechanism of cerebro-cardiac syndrome. Methods The change of dynamic expression of ET mRNA in intracerebral hemorrhage region, myocardium and the special areas of central cardiovascular regulation including hypothalamus, brain stem and hippocampi in intracerebral hemorrhage rats, was detected by in situ hybridization histochemistry (ISHH) technique. Positive reactants were analyzed by the method of quantitative image analysis. Results The quick expression of ET mRNA in the above areas could be evoked because of the introcerebral hemorrhage. The expression of ET mRNA increased at 6 h after introcerebral hemorrhage, peaked at 24 h and decreased until 72 h, but it was still above the normal level. The expression of ET mRNA in myocardium rose simultaneously. Conclusion The excessive expression of ET mRNA in intracerebral hemorrhage region, myocardium and the special central cardiovascular regulating areas may be one of the pathologic bases of cerebro-cardiac syndrome.
