人初始CD_4^+CD_(25)^+调节性T细胞的功能分析

摘要 目的:分离人CD4+CD25+Treg细胞,并进行功能分析。方法:应用免疫磁珠分离正常人外周血中CD4+CD25+Treg细胞,流式细胞仪检测细胞内因子IL-4、IFN-γ和IL-10表达,与CD4+CD25-T细胞共同培养,加或不加入中和抗体抗IL-10和抗TGF-β,检测其抑制功能。结果:CD4+CD25+Treg细胞约占正常外周血中CD4+T细胞的6%(2.4% ̄18.9%,n=15),主要合成IL-10,能够以依赖剂量和非依赖IL-10和TGF-β方式抑制CD4+CD25-T细胞的增殖。结论:CD4+CD25+Treg细胞是一群具有免疫抑制功能的T细胞亚群细胞。 Objective: To isolate and analyse the function of human CD4^+CD25^+ T regulatory (Treg) cells . Methods: Magnetic microbeads were adopted to isolate human CD4^+ CD25^+ Treg cells. The levels of IL-4,IFN-Y and IL-10 were detected by flow cytometry. To assess the properties, CD4^+ CD25^+ Treg cells were cultured with CD4^+ CD25-T cells either in the presence of neutralizing anti-human IL-10 or anti-human TGF-μ antibodies. Results: CD4^+CD25^+ Treg cells accounted for about 6% (2.4%-18.9%, n = 15) of CD4^+ T cells in the peripheral blood of the healthy people. These cells mainly synthesized IL-10,and suppressed the proliferation of CD4^+ CD25-T cells in a dose-dependent and IL-10 and TGF-μ independent manner. Conclusion: CD4^+ CD25^+ Treg cells are a subset of CD4^+ T cells with immunosuppressive function.
