摘要
用放射免疫分析法和比浊法测定API_(0134)对大鼠血浆血栓素B_2(TXB_2)浓度、血小板TXB_2生成血小板聚集的影响。静脉注射API_(0134)70mg或100mg·kg ̄(-1)显著降低血浆TXB_2浓度,其降低率分别为38.8%和51.6%, API_(0134)明显抑制ADP诱导的大鼠血小板聚集和TXB_2生成,抑制率分别为27.8%、39.5%和41.4%、53.6%,两抑制率间呈显著正相关。对胶原诱导的大鼠血小板聚集和TXB_2生成也有明显抑制作用,其抑制率分别为24.3%、35.0%和44.8%、45.5%,两抑制率间相关显著。
Platelet aggregation and thromboxane B_2(TXB_2)production induced by adenosine diphosphate(ADP)or coliagen(Coll),and plasma TXB_2 concentration were studiedon rats by turbidimetry and radioimmunoassay. API_(0134) 70 and 100mg/kg administed iv de-creased d rats plasma TXB_2 cmcentration,the decrements by API_(0134) were 38.8 % and 51.5%respectively. API_(0134) 70 and 100mg/kg injected iv inhibited platelet aggregation as well asthe production of TXB_2, The inhibition were dose-depended;27.8%~39.5%(ADP-induced)and 24.3~35.0%(Coll-induced ) for piatekt aggregation,41.4%~53.6(ADP-indueed)and44.8%~45.5%(Coll-induced)for the TXB_2 production. There was a positive correlation be-tween the inhibition of platelet aggregation and production of TXB_2 induced bv ADP orColl.
