摘要
为经瘤内注射(IT)途径进行肿瘤放射免疫治疗提供组织学实验依据,在裸鼠LS174T移植瘤生长至~1cm时,瘤内分别注射^(125)I标记的小鼠抗人结肠癌单抗(CL-3)和正常小鼠IgG(NMIgG)或腹腔注射(IP)CL-3,3天后取出肿瘤并制备连续切片,用放射自显影术观察^(125)I标记抗体的分布。累加标记抗体在连续切片中每片所占面积,计算总的分布量。结果表明:CL-3 IT组的分布不同于NMIgG IT和CL-3 IP组,CL-3 IT组的标记抗体不仅在注射局部浓度高,而且还通过间质和包膜下扩散到肿瘤细胞,呈成簇分布,但间质中很少;CL-3 IP组仅分布于近间质的3~5层细胞;而NMIgG IT组仅在间质中分布。在瘤内的总分布,CL-3 IT组为2.60±2.19mm^3,是CL-3 IP组(0.08±0.04mm^3)的32倍,是NMIgG IT组(0.37±0.09mm^3)的7倍。CL-3 IT组的肿瘤坏死体积是0.116±0.129mm^3。因此,瘤内注射标记抗体可产生比腹腔给药更好的杀伤肿瘤作用。
PURPOSE To provide a histological basis for radioimmunotherapy by intratumoral administration of labelled antibody. METHODS After the xenografted tumor cell line LS174T grew up to around 1cm in diameter, ^(125)I labelled routine McAb CL-3 and normal mice IgG (NMIgG) were injected intratumorally (CL-3 IT and NMIgG IT) or intraperitoneally (CL-3 IP). Three days later, serial sections of tumor were carried out and distribution was investigated by autoradiography, the total distribution area was calculated by adding the distribution area of radiolabelled antibody in each slide. RESULTS 1) The distribution of CL-3 IT was different from that of NMIgG IT and CL-3 IP. In CL-3 IT group, the radiolabelled antibody not only concentrated in the injection site, but also diffused through interstitial tissues and subeapsular area to tumor cells as clusters with very few in interstitial tissues. In CL-3 IP group, the radiolahelled antibody was only located in three to five layers of cancer cells surrounding some areas of interstitial blood vessels. In NMIgG IT group, the radiolabelled antibody was only located in the interstitial tissues. 2) The average total distribution area of CL-3 IT was 2.60±2.19ram3, it was 32 times of the total distribution area of CL-3 IP (0.08±0.04mm^3), and 7 times of the total distribution area of NMIgG IT (0.37±0.09mm^3). 3) The necrostic area of cancer cell in CL-3 IT group was 0.116±0.129mm^3. CONCLUSIONS Intratumoral injection of radiolabelled antibody may produce better tumoricidal effect than the intraperitoneal administration.
