摘要
结合形态与分子生物学特征对从河北省分离的40个异小杆类昆虫病原线虫种群进行鉴定,其主要形态特征(4个典型种群平均值):侵染性幼虫体长=550(520~610)μm,体宽=22.2(20~24)μm,尾长=85.7(80~88)μm,E%=112.5%(105%~119%);雄虫交合刺长度=36.8(33.75~39.0)μm,引带长度=17.9(16.3~20.0)μm,GS%=0.49(0.41~0.59),SW=1.25(1.15~1.43),与嗜菌异小杆线虫Heterorhabditis bacteriophora形态特征相近.经对其中二个典型种群的ITS-rDNA测序,其ITS1-rDNA序列与已知种类H.bacteriophora的ITS1-rDNA序列相差小于1.2%,和H.argentinensis的差异小于1.5%,而与其他种类的差异则高达18.7%以上.综合其形态与分子特征,确定河北省的异小杆类线虫为嗜菌异小杆线虫.
40 Heterorhabditis spp. populations isolated from Hebei province were identified based on the morphological and molecular characters. The main morphometrical characteristics based on the average of four typical populations are as followed: infective juvenile length = 550(520 - 610)μm, tail length = 85.7 (80 - 88)μm, body width = 22.2(20 - 24)μm,E% = 112.5% (105% - 119% );Male spicule = 36.8(33.75 - 39.0)μm, gubemaculum length = 17.9(16.3 - 20.0)μm, GS % = 0.49 (0.41 - 0.59), SW = 1.25 ( 1.15 - 1.43 ), they are mostly close to H. bacteriophora in morphology. The ITS-rDNA of two local populations was sequenced. The ITSI-rDNA sequence differences are less than 1.2% between local populations and H. bacteriophora, less than 1.5 % between local populations and H. argentinensis, but over 18.7 % between local populations and other known species. It is proved that the Heterorhabditis spp. populations isolated from Hebei province are H. bacteriophora based on morphological and molecular characters.
