摘要
目的用高效液相色谱法测定水红花子中芦丁与槲皮苷的含量。方法芦丁的含量采用ShimadzuC_(18)柱(4.6mm×150mm,5μm),流动相为甲醇-0.05%H_3PO_4溶液(42:58),流速1.0mL·min^(-1),检测波长254nm,柱温25℃,用外标法定量;采用ShimadzuC_(18)柱(4.6mm×150mm,5μm),流动相为甲醇-0.2%H_3PO_4溶液(45:55),流速1.0mL·min^(-1),检测波长350nm,柱温室温,用外标法定量,测定槲皮苷的含量。结果芦丁的线性范围0.053~0.53μg,r=0.9999;槲皮苷的线性范围0.40~2.00μg,r=0.9996。结论该方法简便,快速,线性关系良好。
OBJECTIVE To determine the contents of rutin and quercitrin in polygonum orientale by HPLC. METHODS The analysis of rutin was performed on a Shimadzu C18 column (4.6 mm×150 mm,5μm). The mobile phase was MeOH-0.05%H3P04(42:58) with a flow rate of 1.0 mL·min^-1 at 254 nm. Quercitrin was separated on a Shimadzu Cls column(4.6 mm×150 mm,5μm) .The mobile phase was MeOH-0.2% H3P04(45:55) with a flow-rate of 1.0 mL·min^-1 at 350 nm. Both seperation were performed at room temperature. RESULTS The method was simple and had a good linear relationship. The linear range of rutin was 0.053 - 0.53 μg with a good correlation (r) of 0.999 9. The recovery was 96.30% ,RSD was 1.22%. The linear range of quercitrin was 0.40 - 2.00 μg with a good correlation (r) of 0.999 6. The recovery was 97.80% ,RSD was 1.43% .CONCLUSION The method is rapid,accurate and can be used to determine the contents of rutin and quercitrin in Polygonum orientale.
