摘要 目的:研究高糖对血管内皮细胞增殖、凋亡的影响。方法:用高糖(10、20、30、40、50mmol/L)作用培养的人脐静脉血管内皮细胞(human umbilicalveinendothelial cells,HUVEC),第2、4、6、8、10、12、14天,用氚-标记的脱氧胸腺嘧啶核苷(tritium-labelledthymidinedeoxyribose,3H-TdR)掺入法测定每分钟计数(counts perminute,cpm),流式细胞仪检测技术测定内皮细胞的增殖指数、凋亡指数和细胞周期的变化。结果:在高糖作用下,HUVECcpm值显著低于对照组(P<0.001)增殖指数低于对照组(P<0.05),凋亡指数高于对照组(P<0.05)呈明显的剂量依赖关系,随作用浓度增大时间延长更为明显。各高糖组G1期百分率均升高(P<0.01),S期百分率均降低(P<0.05)。结论:高糖使培养的人脐静脉血管内皮细胞凋亡加速,同时抑制了内皮细胞的增殖。根据G1期细胞百分率升高,S期细胞百分率下降的结果,高糖导致部分内皮细胞可能被阻滞在G1/S转变期。 Objective: To investigate the effect on proliferation and apoptosis of vascular endothelial cells exposed to high glucose. Methods: The cultured human umbilical vein endothelial cells (HUVEC) were treated with high glucose at concentrations of 10, 20, 30, 40, 50mmol/L for 2, 4, 6, 8, 10, 12 and 14 days, cpm value was studied by using tritium - labelled thymidine deoxyribose (^3H - TdR) incorporation assay. Flow cytometry was used to detect proliferation index, apoptosis index and cell cycle. Results: Treated with high glucose, the cpm value was significantly reduced ( P 〈 0.001 ), The proliferation index induced by high glucose was much lower than that of the control groups ( P 〈 0.05) which were dependent on the dose and the time course. The apoptosis index of HUVEC in most groups was higher in dose dependent manner compared with that of the control groups ( P 〈 0.05 ). The percent of the cultured cells in G1 phase in all high glucose groups were increased and that of the S phase was greatly decreased ( P 〈 0.05) in a dose - time dependent manner. Conclusion: Exposed to high glucose, the apoptosis of HUVEC was accelerated and the suppressive effect of high glucose on the proliferation of HUVEC was observed. From the fact that the cells in G1 phase were increased but in S phase were decreased, the endothelial cells were possibly arrested in G1/S checkpoint was suggested.
