P物质在大鼠脑室管膜中定位分布的免疫组织化学研究 被引量:3

A HISTOCHEMICAL STUDY OF LOCALIZATION OF SUBSTANCE P IN THE EPENDYMA OF THE RAT

摘要 SD雄性大鼠侧脑室注射秋水仙素100g/15L,24h后灌注固定,同时经侧脑室注射微量固定液加强对室管膜的固定。用免疫组化ABC法研究P物质在SD雄性大鼠脑室管膜中的分布。结果显示,SP免疫阳性细胞分布于第三脑室侧壁、中脑导水管和延髓中央管的室管膜中,其中位于下丘脑结节区第三脑室侧壁及最后区平面延髓中央管的阳性室管膜细胞为伸长细胞,发出基突伸向周围灰质。本研究首次证实了含SP伸长细胞的存在,结合本实验室其他实验结果提示,这些伸长细胞在含肽的种类和区域分布上存在差异,为进一步深入研究伸长细胞在脑-脑脊液神经体液回路中的重要作用提供了化学神经解剖学依据。 Immunohistochemical ABC technique was applied to study whether and how the neuropeptide,substance P(SP)existed in the ependyma of the rat.The results showed that there were some ependymocytes which were positive by SP immunoreactive in the wall of the third ventricle(V3),the aqueduct of midbrain(MA)and the central canal(CC)of medulla oblongata. These cells belonged to tanycytes and situated only in the ependyma of V3(hypothalamic tuberal area)and CC of medulla oblongata(area postrema),the former tanycytes were ovoid or pyramid with short basal processes into the subependyma,the latter situated more in the dorsal part of CC with cuboid shape and less in lateral and ventral parts of CC with ovoid shape. Their long basal processes extended into the central gray of medulla oblongata which were near the nucleus of the solitary tract,dorsal moter nucleus of vagus,and the hypoglossal nucleus. Since hypothalamus and area postrema are important centers in regulating the motions of internal organs,endocrine and emotions,our results have afforded a new direct evidence which suggests that tanycytes with some neuropeptides may be a new way to communicate the neuroregulation with the humoral regulation,which may be important in physiology.There are many questions on tanycytes remain further studied.
