摘要
目的检测免疫增强因子 Hsp27、Hsp70、Hsp90、IL-2、TNFα及免疫抑制因子 IL-10、TGFβ_1在鼻咽癌中的表达,探讨其在免疫治疗上的意义。方法应用 Western blot 检测 Hsp27、Hsp70、Hsp90在鼻咽癌及鼻咽炎症活检组织中的表达,用双抗体夹心酶联免疫吸附测定(enzyme linked immuno sorbent assay,ELISA)法检测鼻咽癌、鼻咽炎症及正常人血清中 IL-2、TNFα、IL-10、TGFβ_1的表达水平,了解机体的免疫状态。结果鼻咽癌组织中Hsp27较鼻咽炎症组织高表达(P<0.05),Hsp70、Hsp90表达无明显差异,鼻咽癌血清中 TNFα(P<0.01)、TGFβ_1(P<0.01)水平较正常及鼻咽炎症升高,IL-10表达明显降低(P<0.01),IL-2表达无明显变化(P=0.425)。结论根据鼻咽癌患者免疫增强及抑制因子表达情况,可以应用小剂量的 IL-2,TGFβ单克隆抗体,Hsp 多肽复合物疫苗来辅助鼻咽癌的治疗。
Purpose To detect the expression of immune protective factor Hsp27, Hsp70, Hsp90, IL-2, TNFα and immune inhibitor IL-10,TGFβ1 in nasopharyngeal carcinoma (NPC) as well as to explore the potential efficacy on immune therapy. Methods The expression of Hsp27, Hsp70, HspgO in tumor and chronic nasopharyngitis tissue was studied with Western blot method. Serum level of IL-2 ,IL-10,TNFα,TGFβ1 was measured using ELISA. Immune condition of NPC patients was analysed. Results Hsp27 elevated remarkably ( P 〈 0.05 ) while HspgO and Hsp70 showed no significant difference between NPC and control tissues. Expression of serum TNFα,TGFβ1 improved highly wheras IL-10 decreased significantly in NPC compared with normal control (P 〈 0.01 ), There is no alteration in the level of IL-2. Conclusion According to the detection results of immune protective factors and immune iubibitors, We can use little dose IL-2, TGFβ antigen, Hsppolypeptide compound vaccine in patients with nasopharyngeal carcinoma to improve therapeutic effects.
