心肌营养素-1上调大鼠心肌转录因子GATA4 mRNA的表达

摘要 [目的]研究心肌营养素-1(cardiotrophin-1,CT-1)对心肌转录因子GATA4 mRNA表达的影响。[方法]应用RT-PCR半定量技术,分别测定不同刺激剂量CT-1作用24 h及0.1nmol/L的CT-1作用不同时间后,大鼠心肌细胞GATA4 mRNA表达情况。[结果]心肌细胞GATA4 mRNA表达值在0.001 nmol/L、0.01 nmol/L、0.1 nmol/L1、nmol/L及10 nmol/L的CT-1作用24 h后分别为0.504±0.044、0.73±0.029、1.217±0.023、2.109±0.11、2.613±0.089;与对照组0.445±0.024相比,0.01nmol/L以上组均差异显著(P<0.05);在0.1 nmol/L的CT-1作用10 min、30 min1、h、2 h3、h、6 h1、2 h及24 h后分别为0.591±0.2、0.613±0.096、0.618±0.088、0.608±0.132、0.657±0.055、1.109±0.089、1.309±0.111、1.323±0.074;与对照组0.55±0.1067相比,6 h以后均表现为极显著性差异(P<0.01)。[结论]CT-1刺激与大鼠心肌转录因子GATA4 mRNA表达有明显的相关性。 [ Objective] To investigate the effect of cardiotrophin- 1 (CT - 1) on the expression of GATA4 mRNA of cardiomyocytes. [ Methods] Using semi- quantitative RT - PCR, to measure the GATA4 mRNA value of the cultured rat's cardiomyocytes after being treated by CT - 1 with different concentrations for 24 h, or with 0.1 nmol/L for different times. [Results] After being treated with 0.001nmol/L, 0.01 nmol/L, 0.1 nmol/L, 1 nmol/L and 10 nmol/L CT- 1 for 24 h, the results of GATA4 mRNA were 0.504±0.044, 0.73 ±0.029, 1.217±0.023, 2. 109± 0.11 and 2.613±0.089 respectively, there is significent difference between 0.01 nmol/L or higher concentrations and the blank group (P 〈0.01). Treated with 0.1 nmol/L CT - 1 for different times 10 min, 30 min, 1 h, 2 h, 3 h, 6 h, 12 h and 24 h respectively, the results of every groups were0. 591±0.2, 0.613±0.096, 0.618±0.088, 0.608±0.132, 0.657±0.055, 1.109±0.089,1.309±0.111 and 1. 323± 0. 074 respectively. There is no significent difference ( P 〉 0.05) between the control and every one of the former five groups, but it increases significently from 6 h (P 〈 0.01). [ Conclusion] CT - 1 has a significant role on expression of GATA4 mRNA of cardiomyocytes.
