摘要
通过实验研究确定了虎杖活性成分的提取和分离条件:用质量8倍于虎杖粗粉,浓度为80%乙醇,温度60 ℃,浸渍3次,每次2 h,提取率为96.3%.再经硅胶柱层析分离虎杖乙醇的提取物,氯仿-甲醇梯度洗脱[V(氯仿):V(甲醇)=9.8 : 0.2 ~ 7.0 : 3.0].薄层层析检测得到5种单体:大黄素甲醚、大黄素、大黄酸、白藜芦醇和白藜芦醇苷.以虎杖饮片为基准,得率分别为0.03%、0.92%、1.56%、2.0%和1.41%.
This paper reports the experiments of extracting and separation active ingredientsfrom Polygonurn cuspidaturn. The powder of Polygonurn cuspidaturn is steeped in the ethanol with 80% in concentration and 8 times in weight, under 60 ℃ for three times, 2 hours each. The active components with 96.3 in percentage are extracted. Then, these active components are separated on a silica col- umn eluted by chloroform-methanol (ratio of chloroform over methanol about 9. 8 ; 0. 2 to 7. 0 : 3. 0). Five kinds of monomers are identified by TLC to be physcion, emodin, rhein, resveratrol, and piceid, with yield, based on the Polygonum cuspidaturn, 0.03 %, 0.92 %, 1.56%, 2.0 %, and 1.41% respectively.
