摘要 目的:了解早期显性梅毒患者外周血中白介素(IL)-4、、IL-12和干扰素(IFN)-γ的动态表达与预后的关系。方法:于治疗前及治疗后1、3、6、9、12个月分别取患者外周血单一核细胞(PBMC)与梅毒螺旋体(TP)共同培养,分别用ELISA及逆转录(RT)-PCR方法检测培养细胞上清和PBMC中的IL-4、12、IFN-γ及其mRNA的表达,同时观察快速血浆反应素(RPR)试验转阴情况。结果:IL-12和IFN-γ表达水平与血清RPR试验转阴无明显相关性,IL-4表达水平与血清RPR试验转阴率呈负相关。结论:早期梅毒患者外周血IL-4的表达水平可能可以作为判断梅毒预后的一个较敏感指标。 Objective: To investigate the expression of IL-4, 12, IFN-γ in early symptomatic syphilitic patients, and to study its relation with the prognosis. Methods: Peripheral blood mononuclear cells (PBMC) of 33 syphilitic patients at early stage were collected before treatment and 1, 3, 6, 9, 12 months after treatment. After mixed culture of Treponema pallidum (TP)and PBMC, IL-4, IL-12, IFN-γ in supernatant were measured by means of ELISA, and mRNA expression in PBMC by means of RT-PCR. PHA served as positive control and complete 1640 cell culture as negative control. Serum RPR test response of these patients was obtained 1, 3, 6, 9, and 12 months after routine treatment. Results: The level of IL-12 and IFN-γ and their mRNA espression were not correlated with the rate of RPR turning negative. IL-4 level and its mRNA expression were negatively correlated with the rate of RPR turning negative. Conclusions: IL-4 level in peripheral blood of syphilitic patients can serve as a sensitive indicator for the prognosis.
