摘要
通过肾癌细胞的体外实验发现RCC-925肾癌细胞系对长春新碱高度耐药,对阿霉素低度耐药,半数致死浓度分别为14726.4mg/L和1.559mg/L。提示RCC-925细胞具有天然多重耐药性。而环孢素A可显著提高长春新碱和阿霉素的细胞毒性,增效幅度随环孢素A浓度的升高而增强,增效倍数对长春新碱为3.0~355.7倍,对阿霉素为5.4~47.7倍。结果提示环孢素A对RCC-925肾癌细胞的多重耐药性具有显著的逆转作用。
With MTT technique,cyclosporin A was used to enhance the cytotoxic effect of vincristine(VCR)and adriamycin(ADM)on renal cell carcinoma RCC-925 cell line in vitro,LC_(50)was 14 726.4mg/L for VCR and 1.55mg/L for ADM suggesting multidrug resistance of RCC-925.Cyclosporin A above 10.0mg/L would significantly enhance the cytotoxicity of VCR and ADM on the cell line and the effect was increased with the dose.The cytotoxic effect might be increased 3.0 to 335.7 times in VCR and 5.4 to 47.7 times in ADM.The result demonstrated that cyclosporin A could dramatically reverse the multidrug resistance of RCC-925 cells.
