摘要 目的:建立高效液相色谱法测定人血中伪麻黄碱的含量的方法.方法:采用Symmetry C8色谱柱(4.6 mm×250 mm,5μm);以甲醇-11.6%醋酸铵(用醋酸调pH 4.0)(6:94)为流动相,流速:0.8 mL·min-1,紫外检测波长:257 nm,柱温37℃.结果:HPLC法测定人血中伪麻黄碱的线性范围为0.5~4.0 mg·L-1,r为0.998 8.日内、日间差异均小于10%,平均回收率为92.2%.结论:本法适用于伪麻黄碱血药浓度测定. OBJECTIVE An HPLC method was established for the determination of pseudoephedrine in human plasma. METHODS The mobile phase was composed of methanol-11.6% ammonium acetate solution (adjusted to pH 4. 0 by acetic acid) (6:94). The flow rate was 0. 8 mL·min^-1. The detection was carried out with a UV detector at 257 nm,Colum temperature was at 37℃. RESULTS The calibration curve of pseudoephedrine was liner between 0. 5 - 4. 04 mg·L^-1(r= 0. 998 8). Absolute recovery was more than 80%, within-day and between-day RSD were less than 10%. CONCLUSION The method is suitable for the determination of pseudoephedrine in plasma.
