RLM-RACE法快速克隆盐角草胆碱单加氧酶cDN A5′末端全序列 被引量:2

Rapid Amplification of 5′ cDNA Ends of Salicornia europaea Choline Monooxygenase by RLM-|RACE

摘要 采用RLM-RACE法,根据已获得的盐角草胆碱单加氧酶基因的部分序列,设计2条特异性嵌套PCR引物,成功地克隆了该基因cDNA 5′末端全序列,测序结果显示该片段包括5′UTR 154个核苷酸和编码区606个核苷酸,编码N端202个氨基酸.Blast P结果表明,该片段编码的蛋白序列与辽宁碱篷及其他藜科植物的胆碱单加氧酶同源性达到85%以上.同时找出了其转录起始位点,即为该序列的第1个碱基g. Two specific nested PCR primers were designed according to the obtained partial sequences of Salicornia europaea Choline Monooxygenase(CMO) cDNA and the full length of 5′ cDNA ends was cloned by RLM-RACE method. The cDNA fragment included 154 bp 5′UTR and 606 bp in coding sequence and 202 amino acids at the N end of code. The results of Blastp showed that the polypeptide shared more than 85% identity with CMOs from Suaeda liaotungensis and other Chenopodiaceae plants. The transcription initiation site, or the first base of the full sequence, was identified at the same time.
