肾上腺切除的肾阳虚大鼠肝组织蛋白质双向电泳图谱的建立与分析 被引量:15

Establishment and Analysis on Two-dimensional Electrophoresis of Liver Proteins of the Kidney-Yang Deficiency Rats

摘要 目的:建立具有高分辨率和稳定性的大鼠肝组织蛋白质组研究的双向电泳图谱,并对其进行差异蛋白质组分析。方法:正常大鼠和肾上腺切除造成的肾阳虚模型大鼠,肝组织匀浆后提取总蛋白,用Cy3或Cy5标记,每一个Cy3标记样品和一个Cy5标记样品与一个Cy2标记的内标等量混合,上样于同一胶中进行电泳分离,经不同波长单色光激发下扫描得到不同样品的蛋白质组图谱。所获得的图谱用DeCyder软件进行分析。结果:在肾阳虚大鼠肝组织,有8个蛋白质表达水平显著增加,9个蛋白质表达水平显著下降。结论:分析所得的17个差异蛋白质可能与肾阳虚疾病有关。 Objective: To establish and analyse two-dimensional difference gel electrophoresis for proteomic study of Kidney-yang deficiency. Methods: The livers from normal rats and adrenalectomized Kidney-yang deficiency rats were homogenized with lysis solution. The extracts were labeled with Cy3 or Cy5 randomly. Each Cy3-labeled sample and CyS-labeled sample was mixed on the same 2-D gel along with a Cy2-labeled mixture of all samples as an internal standard. The gels were respectively imaged by Typhoon 9400 using different emission filters and spot-features were analyzed by DeCyder software in each paired normal/disease comparison. Result: 8 unique proteins that were changing in abundance more than 1.5 folder up while 9 proteins decreased more than 1.5 folder down between kidney-yang deficiency rats and normal rats. Conclusions: These 17 proteins might be involved in the process of kidney-yang deficiency.
