抗精子抗体阳性大鼠精子凋亡及机制的研究 被引量:12

摘要 目的探讨抗精子抗体(AsAb)阳性大鼠精子凋亡率的变化及其可能的机制。方法建立AsAb阳性大鼠动物模型,HE染色观察大鼠附睾组织形态;采用3’-原位末端DNA标记(TUNEL)技术检测大鼠精子的凋亡率;应用化学比色法测定精子匀浆的MDA含量和总抗氧化能力;应用定磷法测定精子匀浆的Na+-K+-ATP酶和Ca2+-ATP酶活性。结果与正常组相比,AsAb阳性组大鼠精子凋亡率和MDA含量显著增加(P<0.01;P<0.01);总抗氧化能力、Na+-K+-ATP酶和Ca2+-ATP酶活性显著降低(P<0.01;P<0.01;P<0.05)。结论AsAb阳性大鼠精子凋亡率增加;可能与氧自由基含量增多、Na+-K+-ATP酶和Ca2+-ATP酶活性降低有关。 Objective To study the sperm apoptosis and mechanism in Antisperm Antibody(AsAb) positive rats. Methods The animal model of AsAb positive rats was established. The sperm apoptotic percentages in rats were tested by in situ 3'end DNA-labeling(TUNEL). Changes of total antioxidant capacity and MDA content and Na^+-K^+ATPase, Ca^2+- ATPase activity of sperm were detected by chemical colorimetry. Results The sperm apoptotic percentage and MDA content in AsAb positive rats increased as compared with the control group (P 〈 0.01; P 〈 0.01 ). Na^+-K^+ATPase activity, Ca^2+- ATPase activity in AsAb positive rats sperm decreased as compared with the control group (P 〈 0.01; P 〈 0.01 ; P 〈 0.05 ). Conclusion The sperm apoptotic percentage in AsAb positive rats was increased. Increasing oxygen free radicals and decreasing Na^+-K^+ATPase, Ca^2+-ATPase activity might be the mechanism of sperm apoptosis.
