骨桥蛋白诱导的黏着斑激酶信号途径参与大肠癌的侵袭与转移 被引量:1

摘要 目的观察并分析大肠癌中骨桥蛋白(OPN)、整合素β3、黏着斑激酶(FAK)和磷脂酰肌醇激酶(PI3K)的表达特点、相互关系以及与大肠癌进展的相关性,试图揭示介导大肠癌侵袭的信号途径。方法采用SABC免疫组织化学方法,检测这4种抗原在14例结肠癌组织和16例直肠癌组织以及相应的正常组织中的表达。结果OPN在正常组织中主要表达于间质,而在大肠癌组织中主要分布于实质,且随Dukes分期的进展,实质中OPN的阳性染色颗粒逐渐增多,间质中却明显减少;整合素β3在正常组织中呈中等量表达,而在肿瘤组织中几乎检测不到其存在;FAK和PI3K在正常组织中基本无表达,而在肿瘤组织中其表达量明显升高,其表达水平与肿瘤Dukes分期密切相关,二者在肿瘤实质中的表达模式与OPN具有明显的一致性。结论OPN诱导的FAK和PI3K信号途径参与了大肠癌中晚期发生的侵袭与转移过程。 Objective To observe and analyze the expression character and correlation of osteopontin (OPN) ,integrin β3, focal adhesion kinase (FAK) and phosphoinostide 3-kinase (PI3K) and their relation to progression of carcinoma of large intestine to reveal the signal pathway of its invasion. Methods The expressions of OPN,integrin β3 ,FAK and PI3K in 14 cases with colon cancer,16 cases with rectal carcinoma and the corresponding normal tissue were determined by SABC immunohistochemical method. Results OPN mainly expressed in mesenchyme of normal tissue,but expressed in parenchyma of large intestine cancer tissue,and the positive dyeing granule in parenchyma gradually increased,those in mesenchyme significantly decreased with progression of Dukes classification. Integrin β3 moderately expressed in normal tissues,but it hardly expressed in carcinoma tissues. FAK and PI3K did not express in normal tissues,but significantly expressed in carcinoma tissues,which closely related to Dukes classification. The expression model of FAK and PI3K had coherence with that of OPN in carcinoma parenchyma. Conclusions FAK and PI3K signal pathway induced by OPN should participate in invasion and metastasis of later carcinoma of large intestine.
