摘要
目的研究环氧合酶-2(cyclooxygenase-2,COX-2)在膀胱移行细胞癌(transitionalcellcarcino-ma,TCC)组织中的表达及其意义。方法采用免疫组化技术,对49例膀胱TCC石蜡切片和18例正常膀胱黏膜组织进行COX-2蛋白检测,对阳性表达率与肿瘤分级及侵袭性间的相关性与TCC分级及侵袭性进行分析。结果COX-2蛋白在膀胱TCC中表达率为63.2%,正常膀胱黏膜无表达(P<0.01)。低分级TCC阳性率36.4%,高分级为92.8%(P<0.05)。表浅TCC阳性表达率38.9%,浸润TCC为77.4%(P<0.05)。结论COX-2在高分级和侵袭性膀胱TCC中阳性表达率显著上升,可能会成为膀胱肿瘤患者恶性度和判断预后的重要生物学指标,并有可能成为治疗人类TCC的作用靶点。
[Objective] To study the expression and significance of cyclooxygenase-2 (COX-2) in human transitional cell carcinoma (TCC) of bladder. [Methods] A total of 49 paraffin-embedded Tcc tissue and 18 normal bladder sample were stain by SABC immunohistochemical method with specific anti-COX-2 antibody. The correlation between the positive expression rates and grades of tumor was analyzed. [Results] Expression rate of cox-2 protein was significantly correlated to the tumor grades and their invasivence, Expression rate of COX-2 protein in TCC was 63.2%, statistically higher than those in the normal samples (P 〈0.01). Expression of cox-2 were dected in 36.4% with Low-grade TCC and 92.8% with high-grade TCC (P 〈0.05), Expression of cox-2 were dected in 38.9% with non-invasive TCC and 77.4% with invasive TCC (P 〈0.05). [Conclusions] The expression of COX-2 is enhanced in high-grade and invasive TCC, might be a biomarker and as a therapeutic target in human TCC.
