摘要
目的探讨发育期大鼠反复惊厥后海马组织一氧化氮(NO)、一氧化氮合酶(NOS)的变化及川芎嗪对其的影响.方法三氟乙醚反复吸入(连续6次,每天1次)制成发育期大鼠惊厥动物模型,观察川芎嗪对反复惊厥后6 h、1 d、3 d、7 d海马组织NO含量、NOS活性及光镜下海马组织病理变化的影响.结果①反复惊厥后6 h、1 d、3 d海马组织NO含量及NOS活性明显升高,与对照组相比有显著性差异(P<0.01),第7天已恢复正常.②反复惊厥后6 h海马神经细胞即有轻度水肿,第1~3天水肿、变性坏死明显,第7天有大量炎性细胞浸润、胶质细胞增生明显.③川芎嗪干预组海马组织病理变化明显改善,NO含量及NOS活性明显低于惊厥组,两组相比在第6 h、1 d、3 d有显著差异(P<0.01).结论反复惊厥发作可导致大鼠海马组织损伤,可能与NO大量生成,NOS活性明显增高有关.川芎嗪能改善惊厥后脑组织病理损伤,其机制可能与降低NO含量及NOS活性有关.
Objective To study the contents of nitric oxide (NO) and nitric oxide synthase (NOS) activation in the hippocampus and the protective effects of Ligustrazine on the brain injury after recurrent seizures induced by the long - term effect of flurothyl in the growing rats. Methods Seizures in the rats were induced by inhaling flurothyl once a day for six days. To observe the effects of Ligustrazine on the contents of NO and NOS activation at 6 h, 1 d, 3 d, 7 d in the hippocampus after recurrent seizures and the neuropatholngical changes in the hippocampus under light microscope. Results ①The contents of NO and NOS activation at 6 h, 1 d, 3 d in the hippecampus after recurrent seizures were much higher than those in control group ( P 〈 0.01 ), but they recovered at 7 d. ②Under light microscope, light edema of nerve cells in the hippocampus were observed at 6 h, and they become more obvious at 1 d, 3 d and accompanied with degeneration and necrosis of nerve cells, the infiltration of many inflammatory cells and neuroglial cells were seen at 7 d. ③In Ligustrazine treated group, neuronal injury in the hippocampus were obviously decreased, the levels of NO and NOS activation at post - seizure 6 h, 1 d, 3 d group were significantly lower than those in seizure group ( P 〈 0.01 ). Conclusions The damage to the hippocampus induced by recurrent seizures has relationship with the increase of NO and NOS. The suppression of the levels of NO and NOS in the hippocampus probably correlated with the protective effects of Ligustrazine on the brain injury of seizures.
