摘要
目的利用人胃癌BGC-823细胞体外培养实验,观察不同含量ω-3多不饱和脂肪酸合成的二十碳五烯酸(eicosapentaenoic acid,EPA)和二十二碳六烯酸(docosahexaenoc acid,DHA)对肿瘤细胞凋亡的影响。方法采用MTT法、凋亡形态学检查和流式细胞术检测肿瘤细胞凋亡情况。结果30和45 mg/L的EPA或DHA可显著降低肿瘤细胞存活率(P<0.01),透射电镜可见细胞凋亡现象,流式细胞检测bcl-2和Gα基因蛋白表达明显下降(P<0.05,P<0.01)。结论ω-3多不饱和脂肪酸可诱导人胃癌BGC-823细胞凋亡,细胞增殖受到抑制。
Objective To assess the effects of ω-3 polyunsaturated fatty acids -- different content of eicosapentaenoic acid(EPA) and docosahexaenoic(DHA) on apoptosis of human gastric BGC- 823 cells in vitro. Methods Apoptosis of the cells was determined by MTT assay, morphology and flow cytometry. Results After treatment with 30 and 45 mg/L of EPA or DHA, the cell viability was markedly decreased (P〈0.001). Apoptosis of the treated cells was observed with transmission electron microscope, bcl-2 and Ga proteins were decreased by detection of flow cytemetry( P〈0.05, P〈0. 001). Conclusion ω-3 polyunsaturated fatty acids can induce apoptosis of human gastric BGC-823 cells and inhibit the growth of the cultured cells.
