摘要
目的:探讨新生鼠胰岛的分离和纯化,为体外培养胰岛和胰岛细胞奠定基础。方法:将100只新生Wistar大鼠分为10组,每组10只,用V型胶原酶消化胰腺,用质量分数35%牛血清白蛋白(BSA)和葡聚糖作为纯化剂,分离和纯化胰岛。结果:每组胰岛收获量为(780±23)个,纯度达92%,活性为86%,胰岛的分泌功能良好。结论:联合应用35%BSA和葡聚糖分离纯化新生鼠胰岛是可行的。
Objective: To explore the separation and purification of pancreatic islets in neonatal rats. Methods: One hundred neonatal Wistar rats were randomly divided into 10 groups. The pancreas were digested by collagenase V, islets were purified by combination of dextran and 35% bovine serum albumin(BSA). Results: The mean islet yield of each goup was 780 ±23, purity was 92%, viability was 86%, and the islets had good biological activity. Conclusion: The use of dextran combined with 35 % BSA is a feasible method for isolation of islets in neonatal rats.
