摘要
目的:探讨E-cadherin(E-cad)、整合素α5β1与人结直肠癌临床病理特征的关系。方法:采用免疫组织化学SABC方法,联合检测41例人结直肠癌癌组织和癌旁正常组织中E-cad和整合素α5β1的表达。结果:结直肠癌组织中E-cad的表达较癌旁正常组织减低(P<0.0001);整合素α5β1在癌组织中高表达,在癌旁正常组织低表达(P=0.0001);有淋巴结转移组与无淋巴结转移组相比,E-cad低表达率分别为70.00%(14/20)和28.57%(6/21)(P=0.0080);整合素α5β1高表达率分别为90.00%(18/20)和33.33%(7/21)(P=0.0002)。在Dukes分期各期之间E-cad表达差异无统计学意义(P=0.0806);整合素α5β1表达差异有统计学意义(P=0.0024),Pearson列联系数P=0.51。结论:人结直肠癌组织中E-cad表达减低和缺失,可能导致人结直肠癌淋巴结转移;整合素α5β1有一定表达,可能参与结直肠癌发生、发展及淋巴结转移。
Aim: To investingate the expression and significance of E-cadherin (E-cad) and integrinα5β1 in patients with colorectal carcinoma. Methods: Forty-one specimens of colorectal carcinoma and adjacent colorectal tissue were obtained from 41 patients with colorectal carcinoma. Immunohistochemical staining(SABC method) was performed to detect the expression of E-cad and integrinα5β1. The correlation among the expression of E-cad and integrinα5β1 with lymph node metastasis,and Dukes stages of colorectal carcinoma were analyzed. Results: Compared with the adjacent colorectal tissue, the expression of E-cad was lower (P 〈 0. 0001 ) , and that of integrinα5β1 was higher (P = 0. 000 1 ) in carcinoma tissue. In carcinoma tissue with lymph node metastasis and carcinoma tissue without lymph node metastasis, the lower-expression rates of E-cad were 70.00% and 28.57% ( P = 0.0080) , and the higher-expression rate of integrinα5β1 were 90.00% and 33.33% (P = 0. 000 2 ). There was statistic difference in integrinα5β1 expression among different Dukes stages(P =0.0024 ) of colorectal carcinoma. While there was no statistic difference in E-cad expression amoug different clinical Dukes stages ( P = 0. 080 6 ). Conclusion : Loss or lower expression of E-cad can be detected in colorectal carcinoma tissue,which might result in lymph node metastasis. Higher expression of integrinα5β1 may play an important role in carcinogenesis, progression and lymph node metastasis of colorectal carcinoma.
