摘要
目的研究脊神经根撕脱后Semaphorin3A在脊髓中表达的变化。方法建立大鼠脊神经根撕脱模型;半定量逆转录-聚合酶链反应法检测Semaphorin3A(Sema3A)的表达,原位杂交与荧光免疫组化双染法行组织定位检测。结果脊神经根撕脱后,Sema3A在脊髓中表达减少,伤后3d降至最低点,下降70%(P<0.05),至28d仍保持低水平(P<0.05);Sema3A在脊髓神经元细胞中表达,而不在胶质细胞中表达。结论脊神经根性撕脱后,Sema3A在脊髓运动神经元中表达的下调是与创伤相关的适应性反应之一,提示Sema3A的表达可能受远端神经或肌肉细胞成分的逆行性信号调控。
Objective To investigate changes of Semaphorin3A expression in the spinal cord after spinal root avulsion. Methods Rat spinal root avulsion model was established to monitor expression of Semaphorin3A RNA by semi-quantitative reverse transcription and polymerase chain reaction analysis. Double staining by in situ hybridization and fluorescent immunohistochemistry was carried out to detect Semaphorin3A mRNA in the spinal cord tissue. Results The expression of Semaphorin3A mRNA rapidly decereased after spinal root avulsion, reaching its lowest level three days after injury (decreased by 70% , P 〈 0.05) , and remained still low even at day 28 ( P 〈 0.05). Semaphorin3A was expressed in NeuN-positive neurons rather than in glia in the spinal cord, Conclusion The down-regulated expression of Semaphorin3A in motoneurons of the rat spinal cord after spinal root avulsion is part of the adaptive response related to trauma, as implies that the expression of Semaphorin3A mRNA may be regulated by retrograde signals derived from cellular components in the distal nerve or the muscles.
