摘要
作者用家蚕丝素制成丝素溶液,加入大肠杆菌细胞混匀凝固后,再用戊二醛固定和交联,制成固定化细胞丝素凝胶,用于转化延胡索酸为L-天门冬氨酸。测定了固定化细胞转化率和转化效率。实验结果表明,通过控制底物的流速,可使转化率达到95%以上。在转化率为95%时,达到最大转化效率,产率为6.31 g/h。连续使用一个月转化原活力仍保持在92%以上。
Silk fibroin is linked and immobilized by Glutaraldehyde in the solution and made into silk fibrion gels, then applied to the immobilization of the E.coli cells , the immobilized cells was then used to produce L-Aspartic acid from Yanhusuo acid . The radio and efficiency of the transformation have been detected, it is possible to make the ratio of transformation above 95% by controlling the velocity flow. Under this ratio, the efficiency reaches the highest and the yield reached about 6.31g/h. After continuously running of this programe about one month, its enzyme activity remains over 92%.
