摘要
目的探讨组织因子表达对结直肠癌细胞血行转移能力的影响。方法利用构建有正义/反义组织因子cDNA(TFcDNA)的真核表达载体pcDNA3.1/Zeo,以脂质体法转染LOVO细胞,采用Western印迹分析检测转染成功LOVO细胞的组织因子蛋白表达水平。18只裸鼠(Babl/cnu/nu)随机分成3组,分别将转染了正义/反义TFcDNA的LOVO细胞及未转染的LOVO细胞通过尾静脉接种至裸鼠体内。8周后观察裸鼠发生肺转移的数目,以肺转移的数目反映LOVO细胞的转移能力。结果转染了正义TFcDNA的LOVO细胞的组织因子表达水平及肺转移数目较未转染的LOVO细胞升高(P<0.05,P<0.01);转染了反义TFcDNA的LOVO细胞的组织因子表达水平及肺转移数目较未转染的LOVO细胞相比则降低(P<0.05,P<0.01)。结论组织因子可以增加人类结直肠癌细胞的体内血行转移能力。
Objective To investigate the role of tissue factor(TF) in hematogenous metastasis of human colorectal carcinoma cells (LOVO) in vivo. Methods The eukaryotic expression vectors pcDNA3. 1/Zeo bearing either sense or antisense TFc DNA were transfected into LOVO cells by lipofactamine 2000. TF protein expression in the transfected cells was detected by Western blot. Eighteen nude mice (Babl/c nu/nu) were randomly divided into three groups, and then transfected and untransfected LOVO cells were implanted via tail vein respectively. The nude mice were sacrificed 8 weeks after implantation, and the number of metastatic nodules in the lung was used to assess the metastatic ability of LOVO cells. Results Compared with the untransfected group, TF expression of LOVO cells and the numbers of metastastic nodules in the lung increased in sense-TF cDNA transfection group ( P 〈 0. 05, P 〈 0.01, respectively), whereas decreased in antisense-TF cDNA transfection group ( P 〈 0.05, P 〈 0.01, respectively) . Conclusion TF can increase the hematogenous metastatic ability of human colorectal carcinoma cells (LOVO) in vivo.
