摘要
目的建立地高辛血浓度的非放射性检测方法。方法利用进口发光试剂和自制的兔抗地高辛 IgG,建立了化学发光酶免疫测定血清地高辛浓度的方法。结果最低检测限为0.056μg/L;平均回收率98.7%;批内变异系数≤8.8%;临床常用甾体药物无明显干扰;与荧光偏振免疫法相关良好。结论化学发光酶免疫法灵敏、准确、稳定、特异,可以作为地高辛血药浓度监测的非放射性常规方法。
Objective It is to establish a non-radioactive method to measure digoxin serum concen- tration.Methods By utilizing imported chemiluminescent reagent and self-prepared rabbit anti-digoxin IgG,a method of chemiluminescent enzymimmunoassay(CLEIA)was established then evaluated. Results The CLEIA has a lower detecting limit of 0.056 μg/L,an average recovery of 98.7%,an at- the-most whithin-run CV of 8.8% and a remarkably good correlation with Abbott's FPIA(fluorescent polarization immunoassay; r=0. 9725).No significant interference by some patients taken other steroid drugs was detected.Conclusions CLEIA is a sensitive,acurate,stable and specific method:It is a qualified candidate for digoxin serum level monitoring,non-radioactive routine.
