摘要
[目的]探讨性成熟前期褪黑素(melatonin,MT)分泌不足所诱导的神经、内分泌变化与肾阴虚证的关系。[方法] 将30只离乳后的SD大鼠随机分为正常组、模型组、滋肾阴清相火方药组(中药组);将模型组和中药组大鼠24 h/d连续光照30 d,中药组大鼠同时给予滋肾阴清相火方药(13.3 g·kg-1·d-1)灌胃,30 d后于夜间(0:00-2:00)取血,观察大鼠血液中MT、睾酮(testosterone,T)、肾上腺素(epinephrine,E)和去甲肾上腺素(norepinephrine,NE)含量的变化。[结果] 模型组大鼠体质量和血液中MT浓度下降,T、NE、E含量升高,与正常组比较均有显著性差异(P<0.05或P<0.01);给予滋肾阴清相火方药可升高大鼠体质量和血中MT浓度,降低T、NE、E含量,与模型组比较均具有显著性差异(P<0.05 或P<0.01)。[结论]连续光照可减少性成熟前期大鼠血液中MT浓度,同时升高T、NE和E含量,而滋肾阴清相火的中药可使其部分恢复,反证了性成熟前期持续光照导致的MT等分泌不足的改变类似于中医肾阴虚相火旺的证候。
[ Objective] To explore the relationship between neuroendocrine changes caused by melatonin (MT) hytpo-secretion and kidney-yin deficiency syndrome during prepubescence. [ Methods ] Thirty SD rats after ablactation were randomly divided into normal group (group A), model group (group B) and herbal medicine group (group C, treated with kidney-yin-nourishing and deficiency-fire-clearing herbs). Rats in groups B and C were exposed to light continuously for 30 days and simultaneously group C was given kidney-yin-nourishing and deficiency-fire-clearing herbal medicine (13.3 g·kg^-1·d^- 1, qd) for 30 days . After treatment, blood samples were collected at 0 : 00-2 : 00 o'clock to observe the changes of blood MT, testosterone (T), epinephrine (E) and norepinephrine (NE) levels. [Results] Body weight and blood MT level decreased, and T, NE and E levels increased in group A (P 〈 0.05 or P 〈 0.01 compared with group B). Kidney-fin-nourishing and deficiency-fire-clearing herbal medicine increased body weight and blood MT level, and decreased T, NE and E levels (P 〈0.05 or P 〈0.01 compared with group B). [Conciusion] Continuous exposure to light decreases blood MT level, and increases T, NE and E levels in prepubeseent rats; kidney-yin-nourishing and deftciency-fire-clearing herbal medicine can counteract the above changes. This indicates that manifestations in rats with MT hypo-secretion induced by light exposure during prepubescence are similar to the syndrome of kidney-yin deficiency and deficiency-fire hyperactivity.
