摘要
从蜜环菌发酵液提取粗多糖,经分离纯化得到多糖-A和多糖-B两个组分,快速纯化系统(FPLC)和电泳法鉴定各自为均一的组分。FPLC法测定A组分的分子量分别为2.0×105,苯酚-硫酸法测得其总糖含量为86.6%,考马斯亮蓝法测其蛋白含量为12.92%,硫酸-咔唑法测其酸性多糖含量为28%,红外光谱呈现多糖吸收特征峰,含有吡喃糖苷键。β-消去反应初步证明所提取的多糖-A存在O-糖肽键。
A crude polysaccharide could be distilled from Armillaria mellea, after purified by DEAE - Cellulose chromatography, we could get the polysaccharide. Through further purification by Sephacryl S - 300 chromatography, we could get polysaccharide - A and polysaccharide - B which were identified to be homogeneous polysaccharide - protein complex by electrophoresis and fast protein liquid chromatography ( FPLC). The molecular weight of the polysaccharide - protein A was 2.0× 10^5. The content of total saccharide was 86.6%. The content of total protein was 12.92%. The content of total acid polysaccharide was 28%. The IR of polysaccharide A showed characteristic absorptions of polysaccharide. 13 -elimination reaction demonstrated that the saccharide were linked to peptides chain through O -glycosidic linkage in it.
