双低油菜华双3号幼苗全长cDNA文库的构建 被引量:4

摘要 利用RNA5′端模板转换技术,以甘蓝型双低油菜华双3号幼苗为材料,构建了其全长cDNA文库。结果表明:文库的原始滴度为1.25×106pfu·mL-1,重组率为87.3%;扩增后的文库滴度为1.13×109pfu·mL-1。随机挑取12个噬菌斑,利用PCR扩增插入片段,电泳结果显示其长度在0.6~1.5kb之间。 By using switching mechanism at 5′ end of RNA transcript technology, a seedling full-length cDNA library of canola variety Huashuang 3 was constructed. The results indicate that the titer of the unamplified cDNA library is about 1.25 × 10^6 pfu·mL^-1 , the titer of the amplified library is 1.13 ×109 pfu·mL^-1. The recombinant rate of the library is about 87.3%. Twelve clones from the library were chosen randomly for screening cDNA inserts by using PCR, showing that its length of the inserted fragments were from 0. 6 kb to 1.5 kb.
