摘要
目的:观察呋喃二氢吡啶二羧酸酯(EFDP)对血管紧张素II(AngⅡ)诱导血管平滑肌细胞(VSMC)增殖的影响。方法:采用培养的兔VSMC,应用3H-TdR掺入法,观察在AngII促进VSMC增殖过程中,EFDP对VSMC DNA合成的影响及其时间效应。结果:AngII可促进处于静止状态的兔VSMC的DNA合成,36 h时细胞DNA的合成达到高峰。EFDP对AngII诱导的VSMC增殖有显著的抑制作用,并呈现出明显的浓度依赖关系,随着EFDP浓度的升高,VSMC增殖的抑制率也逐渐增加,36 h时,78.40μmol/L的EFDP对VSMC增殖的抑制率为21.37%。结论:EFDP可抑制AngII诱导兔VSMC的增殖,并存在一定的量效依赖关系及时间反应性。
AIM: To investigate effects of diethyl-2,6-diethyl-4-furny-1, 4-dihydropyridine-3, 5- dicarboxylate ( EFDP ) on proliferation of cultured vascular smooth muscle cells ( VSMCs ) stimulated by angiotension Ⅱ( AngⅡ ). METHODS : The rabbit VSMCs were cultured and the inhibitory effect of EFDP on DNA synthesis in vascular smooth muscle cells stimulated by Ang Ⅱ were assayed at different times with different concentrations of EFDP by using ^3H-TdR incorporation in vitro. RESULTS: Ang Ⅱ could significantly promote DNA synthesis in quiescent rabbit VSMCs, with a maximal response at 36th h. EFDP significantly inhibited DNA synthesis of VSMCs in a dose-dependent manner, with a maximal response at a concentration of 78.40μmol/L and at 36th h when the inhibitory rate was 21.37%. CONCLUSION : EFDP significantly inhibits DNA synthesis of ribbit VSMCs induced by AngⅡ in a dose and time dependent manner.
