摘要
目的观察中药复方制剂肾衰宁灌肠液(以下简称肾衰宁,SSN)对体外培养的人肾小球系膜细胞产生细胞外基质成分的影响,冀以从细胞生物学水平探讨肾衰宁防治慢性肾功能衰竭(Chronicrenalfailure,CRF)的作用机制。方法采用双抗夹心ELISA法和放免法,观察SSN大鼠血清对体外培养的人肾小球系膜细胞产生纤维连接蛋白(FN)、层粘连蛋白(LN)和胶原Ⅵ(ColⅣ)的影响。结果肾衰宁灌肠液大鼠血清可抑制体外培养的人肾小球系膜细胞产生FN、LN和ColⅣ,其抑制程度与药物浓度有一定的量效关系,即随药物浓度的增加对系膜细胞产生FN、LN和ColⅣ的抑制率也相应增加。结论肾衰宁对体外培养的人肾小球系膜细胞产生FN、LN和ColⅣ有明显的抑制作用,提示肾衰宁抑制系膜细胞产生细胞外基质是预防肾小球硬化发生发展的重要作用机制之一。
Objective To study the effect of Shenshuaining（SSN）, a traditional chinese herbal medicine, on fibronectin （FN）, laminin （LN） and collagen type Ⅳ （Col Ⅳ）produced by human mesangial cells（huMsC） cultured in vitro ,and explore the mechanism of SSN on prevention and treatment of chronic renal failure（CRF） at level of cell biology. Methods The effects of SSN rat serum on components of glomerular extracellular matrix produced by human mesangial cells（huMsC） cultured in vitro were observed by using. ELISA and radio immunoassay. Results The production of FN,LN and Col Ⅳ was significantly inhibited by SSN rat serum in a dose-dependent manner. Conclusion The inhibition by SSN of components of glomerular extracellular matrix produced by human mesangial cells is one of important mechanism of preventing the development of glomerulosclerosis.
