摘要
目的优选并确立贯叶连翘中抗抑郁成分的最佳提取工艺。方法以总金丝桃素含量、总黄酮含量及浸膏得率为指标,采用正交试验法对提取过程中提取温度(A)、提取时间(B)、乙醇浓度(C)及乙醇用量(D)4个因素进行优选研究。结果影响从贯叶连翘中提取抗抑郁成分的主要因素是乙醇浓度与提取温度,贯叶连翘的最佳提取工艺为A4B2C2D1,即20倍量的70%乙醇,80℃回流提取2h。结论本实验设计合理、测定方法可行,实验结果可靠,为大批量从贯叶连翘提取抗抑郁成分提供了理论依据。
Aim To optimize and establish the extraction technology of antidepressive substances from Hypericum perforatum L. Methods The orthogonal test was conducted to test the effect of the temperature of extraction, time of extraction, concentration and volume of ethanal on extracts yield and the content of hypericins and and total flavonoids from Hypericum perforatum L., and optimize the extraction technology. Results Temperature of extractiont and concentration of ethanal had significant effect on the yield of antidepressive substances from Hypericum perforatum L. The optimum extraction technology was A4B2C2D1 ,that is,the circumfluence extraction with 70% ethanol was 20 times the weight of the sample with 2h at 80℃. Conclusion The test, reasonable in design and credible in findings is easily operated, which can offer credible theories for the batch extraction of antideprsstive substances from Hypericum perforatum L.
