摘要
以Fmoc-Leu-Wang树脂为载体,根据Fmoc方法化学合成血管紧张素Ⅰ(AⅠ)。粗肽经RP-HPLC分离后用电喷雾质谱法测其相对分子质量。结果表明,反相主峰明显,杂质少,电喷雾质谱测得的m/z1296.8与AⅠ的[M+H]+离子理论值(1297)相符。试剂除杂处理很重要,操作技术的改进使方法更实用。
Angiotensin Ⅰ was synthesized using Fmoc chemistry on Fmoc-Leu-Wang resin. The synthetic crude product was separated by reversed phase HPLC. The molecular weight of major component was detected by ESI-mass spectrometry operating in positive ion mode. The results showed that the crude product contained only a major component and several trace-level components. The rn/z of the major component was 1296.8, corresponding to the protonated molecular ion ( [ M + H ]^+ ) of angiotensin Ⅰ. In addition, water should be eliminated from chemical reagents, and some practical methods were recommended to make the experiment convenient.
