溶剂系统对RP-HPLC分离性能的影响

摘要 反相高效液相色谱(RP-HPLC)中所用的溶剂系统是影响其分离性能的重要因素之一。本研究以尿嘧啶、苯酚、萘、芘、联苯、1-萘酚、2-萘酚、邻甲酚为标准样品,考察了溶剂强度、溶剂极性等对反相色谱分离性能的影响。采用溶剂三角形法优化了多组分样品分离的溶剂体系,得出最佳分离溶剂系统为甲醇/水(1∶1,V/V)。在此条件下,对中药茶多酚及大黄提取物进行了分离,效果较好。 Solvent system, which is used in RP-HPLC, is one of the significant factors affecting the separation performance. In this study, uracil, phenol, naphthalene, pyrene, biphenyl, 1-naphthol, 2-naphthol, o-cresol were used as standard sam- ples, and the effects of solvent strength, solvent polarity and other factors on the performance of RP-HPLC were investigated. Using the solvent triangle method, the separation of multi-component samples was optimized, and the optimum solvent system was the mixture of methanol and water (1:1,V/V). Under these conditions, the herbal extracts of tea polyphenols and rhubarb extract were separated, the results were satisfied.
