银杏内酯、4-AP和TEA对AGZY-83-a细胞的作用研究

摘要 目的研究中药银杏内酯A、B及钾离子通道阻滞剂4-氨基吡啶(4-AP)和四乙胺(TEA)对人肺腺癌细胞株AGZY-83-a的抑制作用。方法应用体外培养的人肺腺癌细胞株AGZY-83-a,首先采用MTT法测定不同剂量银杏内酯A、B和4-AP、TEA对肿瘤细胞生长的抑制作用,于酶标仪测OD值(λ=490 nm),计算其抑制率;同时进一步绘制药物作用7天的生长曲线以观察其抗肿瘤作用。结果应用不同剂量银杏内酯A和B 48h后测得的OD值与正常对照组无显著差异,1μmol/L银杏内酯A和B对肿瘤细胞增殖的抑制率分别为5.05%和2.52%(n=6,P>0.05);不同剂量4-AP、TEA对人肺腺癌细胞有明显的抑制作用,呈浓度依赖关系。1mmol/L4-AP和5mmol/L TEA对肿瘤细胞增殖的抑制率分别为15.40%和30.69%(n=6,P<0.001)。不同作用时间的银杏内酯A、B对人肺腺癌细胞生长亦无抑制作用;不同作用时间的4-AP和TEA对人肺腺癌细胞有抑制作用。结论银杏内酯A、B对人肺腺癌细胞株AGZY-83-a的生长无抑制作用。4-AP和TEA对人肺腺癌细胞株AGZY-83-a有明显的抑制作用,与药物浓度正相关。 Objective To observe the pharmacological antitumor effect of ginkgolide A, ginkgolide B and 4-aminopyridine (4-AP) and tetraethylammonium (TEA) of potassium channel blockage on human pulmonary adenocarcinoma AGZY-83-a ceils in vitro. Methods MTT assay was used to detect the pharmacological inhibitory effect of the ginkgolide A, B and 4-AP and TEA with different concentrations on human pulmonary adenocarcinoma AGZY-83-a cells in vitro and OD value(λ= 490 nm) was measured to caculate inhibitory rate of cell proliferation. At the same time, the antitumor activities during the seven days treatment period were evaluated by the cell continuous growth curve. Results The OD values had no significant difference compared with the control group after the administration of different concentrations of ginkgolide A and ginkgolide B. After 48 hours' incubation of 1μmol/L ginkgolide A and B with human pulmonary adenocareinoma AGZY-83-a cells, the inhibitory rate of cell proliferation was 5.05 % and 2.52 %, respectively ( n = 6, P 〉 0.05 ). 4-AP and TEA inhibited the proliferation of AGZY-83-a cells in a concentration dependent manner. The inhibitory rate of cell proliferation of lmmol/L 4-AP and 5mmol/L TEA was 15.40% and 30.69%, respectively ( n = 6, P 〈 0.001 ). No inhibition effect of ginkgolide A and ginkgolide B on human pulmonary adenocarcinoma AGZY-83- a cells was found with different times .4-AP and TEA affected the growth of human pulmonary adenocarcinoma AGZY-83-a cells in different time. Conclusion Ginkgolide A and ginkgolide B have no effect on the growth of human pulmonary adenocarcinoma AGZY-83-a cells. 4-AP and TEA affect the proliferation of the AGZY-83-a cells significantly.
