摘要
鸡组织样品经甲醇- 0.2 mol/L NH4H2PO4溶液提取,正己烷洗涤,C18固相萃取柱净化后,采用高效液相色谱法测定了鸡组织中维吉尼亚霉素M1的残留量.色谱条件为二极管矩阵检测器检测;波长为245 nm;色谱柱为ODS-3C18柱(4.6 mm×250.0 mm,5.0 μm);流动相为乙腈∶水=37.5∶62.5(体积比).该方法对肌肉、肝脏和肾脏等不同鸡组织中3种维吉尼亚霉素M1浓度进行回收测定.结果表明:该方法回收率达74.5%~91.1%;批内、批间变异系数均在10%以内;最低检测限为10 ng/g.
A method was developed for detecting virginiamycin M1 in chicken tissues by HPLC with DAD detection at 245 nm. Virginiamycin M1 was extracted from tissues with methanol and 0. 2 mol/L ammonium dihydrogenphosphate, purified by passing through AccuBOND Ⅱ ODS-C18 solid-phase extraction cartridge. For analysis, a ODS-3C18 (4.6 mm × 150. 0 mm, 5.0μm) was used with water-acetonitrile (62.5: 37.5) as mobile phase. Average recoveries of virginiamycin M~ from chicken tissues fortified at a level of 20 -320 ng/g were 74.5% -91. 1%. Coefficients of variation were below 10%. The detection limit for virginiamycin M1 was 10 ng/g.
