摘要
目的观察脊髓损伤(spinalcordinjury,SCI)后细胞凋亡和相关调控基因(FasL和Caspase3)的表达情况。方法通过建立静压型SCI模型(30g的重量,压迫T10节段10min),并用TUNEL染色(terminaldeoxynucleotidyltransferasemediateddUTPbiotinnickendlabeling)和免疫组化染色方法,了解SpragueDawley大鼠SCI后(伤后6、12、24、48和72h以及伤后7、14、21d)细胞凋亡以及相关调控基因(FasL与Caspase3)表达的情况。结果大鼠SCI后损伤节段(T10节段)6h出现TUNEL阳性细胞数增加,12h达到高峰,持续3天~1周后显著减少;损伤相邻节段(T9、T11节段)伤后12h出现TUNEL阳性细胞,伤后3dTUNEL阳性细胞数达峰值;SCI后FasL与Caspase3表达均有不同程度的增高。结论大鼠脊髓损伤后多节段、长时间存在着大量的细胞凋亡;细胞凋亡可能与FasLCaspase3途径有关。
Objective The purpose of this study was to evaluate the cells apoptosis and the expression of FasL/Fas, Caspase-3 in injured spinal tissue. Methods SCI models were produced by static compression(30g weight compressed 10 minutes at T10 level) in adult Sprague-Dawley rats; Injured spinal cord tissues cells apoptosis were examined by the terminal deoxynucleotidal transferase-mediated dUTP-biotin nick end labeling(TUNEL) reaction at 6, 12,24,48, and 72 hours, and at 1,2, and 3 weeks post-SCI ; FasL and Caspase-3 expression was determined by immunohistochemical analysis at 6,12,24,48,and 72 hours,and at 1,2,and 3 weeks post-SCI. Results At the injury segment (T10 level) ,many Tunel positive cells were present at 6 hours after injury, and at 12 hours after injury the number of Tunel positive cells was to the maximum. Morphology indicated that many of these Tunel positive cells were glial cells. By 3 days and 7 days after injury, the number of Tunel positive cells within the injury segment had fallen sharply. Nearby the injury segment (T9 or Tll level),Tunel positive cells were present at 12 hours after injury and reach its peak at 3 days. The expression of FasL and Caspase-3 were increased after injury accordingly. Conclusion Analysis of the results indicated that apoptosis plays a role in the secondary spinal cord injury;Activation of FasL and Caspase-3 is related to cells apoptosis in the injured spinal cord.
