Apoptosis of the cells from Photofrin-mediated photodynamic therapy for Glioma

摘要 目的研究光动力学治疗前后肿瘤残腔壁组织细胞凋亡情况,观察光动力学治疗效果,以期为进一步改善临床应用奠定基础。方法手术中在切除或大部分切除胶质瘤之后的肿瘤残腔壁,分别于光动力学治疗(PD T)前后在对应点取少量组织,分别对标本行TU N EL法凋亡细胞免疫组化病理检验及H E染色。结果病理切片共29张,光动力学治疗前16张仅3张出现凋亡细胞,细胞凋亡率平均0.05%;光动力学治疗后的13张均出现了阳性染色细胞,细胞凋亡率平均39.16%。结论Photofrin介导光动力学治疗胶质瘤可以诱导肿瘤细胞的快速凋亡。 Objective To study the apoptosis response of glioma cured by Photofrin-mediated photodynamic therapy (PDT) in vivo and evaluate the therapeutic efficacy of this method to improve the clinical application of it, Methods After total or subtotal resection ofglioma under microscope, specimen was got at the brim of the cavity. Immediately after PDT, another specimen was obtained at the same localities of the cavity. The apoptotic cells of these specimens were identified with immunohistochemical method (TUNEL) and the tumor cells with HE staining. Results Totally there were 29 specimens, In the 16 specimens before PDT, 3 expressed DNA breakpoint by TUNEL and the average local incidence of apoptosis is 0.05%. All of the 13 specimens after PDT were found positive by TUNEL and the average local incidence of apoptosis is 39.16%. There are significant different between these two groups (P=0.001, t-test). Conclusion Photofrin-mediated PDT can induce rapid apoptotic response in tumor cells.
