摘要
目的克隆表达型别特异性丙型肝炎病毒HCV 1b、2a嵌合抗原,并用于HCV血清分型检测。方法分别对HCV 1b、2a的C区和NS4区进行表位预测,选取优势表位序列连接后,于PGEX-4T-2载体中克隆表达嵌合的HCV 1b、2a抗原,应用竞争抑制酶联法对44份HCV 1b和18份2a血清进行血清分型检测。结果 62例血清样品中50例能分型,其中1b型34例,2a型16例,其分型率为80.6%(50/62),与基因型的符合率为90.0%(45/50)。结论丙型肝炎病毒HCV 1b、2a嵌合抗原用于HCV的血清分型检测,具有较高的准确率和分型率,可用于HCV血清分型检测。
Objective To clone and express the HCV 1b and 2a genotype-specific antigen for serotyping. MethodsTo carry out HCV 1b, 2a of the C and NS4 epitope prediction respectively. After the sequence connection of donunate epi-topes, to clone and express chimeric HCV 1b, 2a genotype-specific antigen in PGEX-4T-2 vector, and to use competitive in-hibition ELISA method to test 44 sera of genotype 1b and 18 sera of genotype 2a. Results The 50 of 62 serum specimenswere serotyped, 34 sera of serotype 1b and 16 sera of serotype 2a,the sensitivity was 80.6 %(50/62),the concordance be-tween hepatitis C virus serotype and genotype was 90.0%(45/50). Conclusions The data indicates that the HCV 1b and2a genotype-specific antigens used in the HCV serotype have high sensitivity and specificity,and could be used in HCVserotyping.
