紫外线照射充氧自血回输对光气中毒家兔肺脏氧化损伤的保护作用

Protective Effects of Ultraviolet Blood Irradiation and Oxygenation on the Lung Free Radical Injury Caused by Phosgene Intoxication

摘要 目的探讨家兔光气中毒后肺脏氧化损伤情况及紫外线照射充氧自血回输(ultraviolet blood irradiation and oxygenation,UBIO)的保护作用。方法将健康家兔50只随机分为5组,每组10只,包括正常对照组、中毒组、常规治疗组、UBIO治疗组及UBIO联合常规治疗(复合治疗)组,连续观察7 d,记录家兔存活率,检测肺匀浆中总超氧化物歧化酶(T-SOD)活性、还原型谷胱甘肽(GSH)、氧化型谷胱甘肽(GSSG)及丙二醛(MDA)含量。结果家兔光气中毒后肺匀浆中GSH活性明显低于正常对照组(P<0.01),而T-SOD活性及GSSG、MDA明显高于正常对照组(P<0.01);经UBIO治疗或复合治疗后,肺组织中GSH含量及T-SOD活性明显高于中毒组(P<0.01),而GSSG、MDA含量明显低于中毒组(P<0.01)。结论家兔光气中毒后可引起肺脏氧化损伤,UBIO治疗有明显的保护作用,可以用于急性光气中毒的治疗或辅助治疗。 Objective To investigate the lung free radical injury induced by acute phosgene intoxication in rabbits and the protective effects of ultraviolet blood irradiation and oxygenation(UBIO).Methods Fifty healthy rabbits were randomly divided into five groups,including normal control,intoxication,routine therapy,UBIO therapy and UBIO therapy combined routine therapy(combined therapy) group.Having been investigated continuously for 7 d,the livability of rabbits was recorded.The activity of superoxide dismutase(SOD),contents of glutathione(GSH),oxided glutathione(GSSG) and malondiadehyde(MDA) in lung tissue were determined respectively.Results The activities of lung GSH in intoxication group were lower than those of control group(P<0.01),but the contents of SOD,GSSG and MDA were higher than those of control group(P<0.01).After UBIO therapy or combined therapy,the content of GSH and the activity of SOD were higher than that of intoxication group(P<0.01),but contents of GSSG and MDA was lower than those of intoxication group(P<0.01).Conclusions Phosgene intoxication can induce lung free radical injury in rabbits,UBIO therapy had obviously protective effect,and it can be used as a treatment or an adjuvant treatment for acute phosgene intoxication.
